Was just wondering if you have read/write on a mesh active and create a prefab from that mesh will deselecting read/write on the original mesh change the prefab as well?

I am going to bet that changing the FBX import setting mesh read/write setting does not require changing the prefabs.
Here’s why: when you use that mesh, that usage is simply a link. It doesn’t copy that particular data over.
Based on that I speculate that there is only one copy of that imported mesh data, and that mesh is marked however you imported it.
But hey, try it out! Let us know. Couldn’t take you more than five or ten minutes…
